What is mtDNA?
Mitochondrial DNA (mtDNA) is a unique form of genetic material found within cellular mitochondria, the “powerhouses” that provide energy. Unlike nuclear DNA, which is inherited from both parents, mtDNA is passed down exclusively from mothers to their children. This unique inheritance pattern makes mtDNA ideal for tracing your maternal ancestry across generations.
What is mtDNA Sequencing?
mtDNA Sequencing is a method used to read and analyze the sequence of nucleotides in the mitochondrial DNA (mtDNA). The mitochondrial genome is relatively small and circular, and it consists of three main regions: two hypervariable regions (HVR1 and HVR2) and a coding region:
- HVR1 and HVR2: These regions are known for their high mutation rate, which makes them particularly useful for studying human migration patterns, tracing maternal lineage, and identifying genetic relationships between individuals. The hypervariable regions contain about 1,100 bases (nucleotides) of the mtDNA genome, a small fraction of the total genome, but significant in terms of genetic variation.
- Coding Region: This part of the mtDNA genome contains genes essential for mitochondrial function, including those involved in the production of proteins necessary for the mitochondria’s energy-generating process. The coding region encompasses the majority of the mitochondrial genome outside the hypervariable regions.

Which test option should I choose?
The Standard (HVR1) Test is a good starting point for most people seeking to learn about their basic maternal lineage and matches. However, the Advanced and Premium options provide additional details:
Advanced (HVR1 + HVR2) – Recommended if you want full access to our ancestry databases to find the closest population matches and potential living maternal relatives.
Premium (Full Sequence) – The highest level providing the complete mtDNA sequence. Required to definitively confirm your precise maternal haplogroup and subclades.
How is mtDNA inherited and passed down?
Only females can pass mtDNA to their children. While males receive mtDNA from their mothers, they do not pass it to any offspring. This allows mtDNA to trace an unbroken maternal line back through many generations.
